A diverse and complex symbiotic microbial community colonize in human host,espicially in the gastrointestinal tract,refer as gut microbiota.bacteroides spp,that include gut microbiota have significant roles on human physiology and metabolism.in present study we evaluate the efficacy of saturated and unsaturated fatty acids on the growth and outer membrane vesicle production of b.fragilis and b.thetaiotaomicron
Methods
B. fragilis atcc 23745 and b. thetaiotaomicron atcc 10774 were grown on bhi broth supplemented with hemin and menadione to harvest massive biomass. from palmitic acid and palmitoleic acids stock with concentration of 20mg/ml were inoculated to culture media with dilution 0,125,250 and 500mg/l.by usind multiple centrifugation and tris-ethylene diamine tetra acetoc acid (edta),sodium deoxycholate buffer bacterial omv were extracted.
Results
Under same condition,both b.fragilis and b.thetaiotaomicron had a significant growth in the culture medium containing palmitic acid than the fatty acid-free medium. furthermore,the b.thetaitomicron
in the medium containing palmitoleic acid had a relatively higher growth rate,while the presence of palmitoleic acid in the culture medium didn't have much effect on growth of b.fragilis. in the case of omv,b.fragilis produce more omv in the presence of palmitic acid,whereas for b.thataitomicron under the same condition,no particular change was observed. in contrast,the production of omv derived from b.thetaitomicron in the presence of palmitoleic increased.
Conclusion
Considering the importance of gut microbiota and their omv in human health, knowing the effect of fatty acids on the growth and production of their omv,fatty acids can be used as their stimulant for the human immune system as well asmetabolism
