The complex series of cellular signaling molecules and transcriptional factors are involved in differentiation and development of mammalian male sexual gonads, testes. several critical genes are involved in this period of gonadogenesis and the regulated genetic network is the basis of this molecular pathway, generally. so the special antibodies that might be detected these genes are one of the purpose of many recent researches.
Methods
The preparation of a monoclonal antibody against testis antigens, 10c3 mab, was performed by hybridoma technique in the our previouse study. for the localization of produced antibody on the embryonic development of mouse was done on the mouse embryo, e8.5 and e14.5, by immunohistochemistry approach. for this purpose, the sagital slices were prepared from different parts of e8.5 and e14.5 mouse embryos. in the immunohistochemistry method, the slides were prepared by using the produced monoclonal antibody, 10c3 mab. the embryonic sections were examined by fluorescence microscopy.
Results
The characterization of produced antibody, 10c3 mab, on the embryonic development only showed that the target genes could be detected on the early embryonic development, e8.5, by 10c3 mab.
Conclusion
The prepared antibody, 10c3 mab, showed high specificity for the proteins that are encoded from critical genes involving in the early development of testis in both embryo and adult mouse testis tissue. in summary, identification of testis development genes (tdg) in mammalians by specific monoclonal antibodies could be helpful in functional characterization of each stage of the development of this sexual glands and could eliminate the occurance of defects in each phases of testis development.
