The influence of lactobacillus fermentum cytoplasmic extract, isolated from traditional cheese (kuzeh) on the expression of p53 gen in k562 cancerous array via a quantitative real time pcr method
The probiotics are able to induce apoptosis in cancerous cells. chronic myelocytic leukemia (cancerous array of k562) is regarded as one of the most prevalent malignancies in human body and its prominent inhibitor gen is p53, which is undergone mutation in more than 50 percents of human cancers. this gen has been famed as "daemon gen" and has the vital role of genetical stabilization and tumor suppression. this gen performs these duties by apoptosis induction, inhibition of cellular cycles, and inhibiting the production of vascular system.
Methods
The aim of the present study is to investigate the influence of cytoplasmic lactobacillus fermentum extract on cellular death in k562 cancerous array and the evaluation of its influence on expression of p53. in this manner, lactobacillus fermentum was firstly cultured in mrs brath, which it was isolated from traditional cheese (kuzeh), and then a sonicator was used to grind the bacteria and its cytoplasmic extract was finally separated. in the next step, different concentrations of this extract was prepared (100, 200, 300, 400,and 500 mg.ml-1) and their anti tumor characteristics against cancerous array of k562 was measured by chromometery (mtt) in laboratory conditions. eventually, a real time pcr method was utilized to investigate the expression of p53.
Results
The results showed that the concentration of 300 mg.ml-1 of the extract caused a significant death induction for cancerous cells (p=0.011). the results obtained from real time pcr investigations also showed that the concentrations of 300 and 500 mg.ml-1 of extract significantly increased the expression value of p53 based on reference group (p=0.05).
Conclusion
: based on the acquired results, it can be concluded that cytoplasmic extract of lactobacillus fermentum, isolated from traditional cheese (kuzeh) has a significant toxicity impact on k562 human eritro miloid cancerous cells. this phenomenon is able to provide a potential technique for targeted treatment of cancer.
