Abstract
Introduction
This article studies the antiapototic effect of cytokines baff & april on the survival of mononuclear cells derived from patients diagnosed with chronic lymphocytic leukemia (cll) and cultured in vitro. cll is endemic in age-old individuals and the death rate among cll patients in the us exceeded 4000 cases in 2014
Methods
Whole blood samples were collected from 25 patients diagnosed with cll and mononuclear cells were separated using ficoll density gradient centrifugation. cytokines baff & april were reconstituted using rpmi. cell viability was measured using trypan blue staining method during cell culture, as well as annexin-v using flowcytometry
Results
The b-cll cells did not show any significant differences in their resistance to apoptosis, when baff & april cytokines added to the mononuclear cell cultures compared to control b-cll cells with no added ones (p>0.05)
Conclusion
Separation of various mononuclear cells and not specifically b-cll cells lead to the environment simulate the condition inside the body. some of those mononuclear cells, so called, "nurse-like cells" (nlcs) had a protective role in the cells' resistance to spontaneous apoptosis. therefore, whether or not we added the cytokines to the culture media, the cells would survive from programmed cell death. this concept led to a new suggestion, that whether adding of anti-baff and anti-april to the culture media of these cells can deprive the cells thoroughly from these cytokines.
