Abstract
Introduction
Nanoparticles have been used for clinical applications such as hyperthermia, increasing the resolution of magnetic resonance imaging and delivering drugs, and also have a great potential for using in tissue repair and treatment of cancer. present study aimed to assess the effects of iron oxide nanoparticles on fetal heart development by using gene expression profiling approach.
Methods
Female syrian mice had been grouped in control, sham and experimental groups. after mice get pregnant, in experiment group, on ninth day of pregnancy they were injected intraperitoneally by the nano iron oxide. in sixteenth day of pregnancy, the embryo removed from mother's body and its heart was isolated. rna extraction from heart tissue and cdna synthesis was conducted and whole genome expression analysis operated by using dna microarray with mouse wg v2.0 expression bead chip. results of affected genes were confirmed by using qpcr.
Results
Results were showed significant down regulations in mrna level of nine genes in heart tissue of treated embryonic mice. five affected genes were member of gap junction proteins and nkx-homodomain transcription factors that are involved in myelination and organs development repeatedly.
Conclusion
It seems that nano ironoxide could affect on gene expression pattern of embryonic heart development and may consider as potential harmful material for pregnant women
