Abstract
Introduction
Thiosemicarbazones and their metal complexes have significant biological activities including anticancer, antibacterial, antiviral, antifungal and antioxidant. the biological properties of thiosemicarbazones are frequently dependent to metal ion coordination. in the present study, the effect of anti-proliferative and oxidative stress –induced apoptosis by the nickel complex of thiosemicarbazone (4-hbtc) compound on the human kg1a leukemia stem cells was examined.
Methods
The kg1a cells were treated with various concentrations (10-150 μm) of the 4-hbtc for 3 days and cell viability was determined by mtt growth inhibition assay. furthermore, antioxidant enzymes superoxide dismutase (sod) and catalase (cat) levels were evaluated.
Results
Our results indicated that the cell proliferation was inhibited in a dose and time dependent manner. the cytotoxic effect of the compound was determined with an ic50 of 100 µm. this compound increased cellular sod and cat activities in 24 and 48 hr and reduced activity of the cat after 72 h exposure in comparison with corresponding control groups.
Conclusion
According to our data, it was shown that 4-hbtc inhibits cell viability of the kg1a cells associated with oxidative stress –induced apoptosis in a time and dose dependent manner. our findings suggested that this compound can be considered as a great candidate for pharmaceutical evaluations.
