molecular detection of factor 5 leiden (g1691a) and its relation with tnf- α gene polymorphism in human abortion

Abstract


Introduction

Abortion is the most common of pregnancy complications. in the pathogenesis of abortion as a multifactorial disease, genetic factors and environmental factors are related. hereditary thrombophilia as one of the factors known to be involved in abortion and factor 5 leiden (g1691a) is the most important factors is the hereditary coagulation thrombosis. another factor in recurrent miscarriage fetal inflammatory factors is considered that tnfα is one of the most important proinflammatory cytokines and played a key role in many infections and inflammatory diseases as well. polymorphism tnfα susceptibility to various diseases associated with chronic inflammation. of course, this polymorphism association with abortion caused by inflammation in iran has been less studied. this study aimed to investigate the relationship between factor 5 leiden (g1691a) and tnfα - 308 with abortion.

Methods

: in this case-control study, a total of 105 women were included in this study: 47 women with two or more consecutive miscarriages and 58 healthy controls. dna extracted from the venous blood and then , g1691a and tnfα -308 genotypes detected by multiplex- pcr and electrophoresis techniques .

Results

The frequency of 1691g/g genotype was 87% in patients and 95% in controls while the frequency of 1691g/a genotype was 13% in patients and 5% in controls. in other hand, the frequencies of tnfα -308 gg, tnfα -308 ga and tnfα -308 aa genotypes were (19%, 45% and 36%) in patients and (54%, 31% and 15%) in controls, respectively. there is significant difference in the prevalence of 1691a/g genotype and -308 aa genotypes among women with patients and healthy controls.

Conclusion

it can be concluded that detection of factor 5 leiden (g1691a) and tnfα - 308 genotypes in early detection and early treatment in abortion is effective.
