9-tbap from spiroaminopyrimidones family decreases cell proliferation and down-regulation of survivin concomitant with induction of apoptosis in nb4 leukemia cells.

Abstract


Introduction

It has been recently reported the activity of aminopyrimidone family to induce apoptosis in human cancer cells. in this study, we reported an active compound from spiroaminopyrimidone family with apoptotic activity against nb4 acute promyelocytic leukemia cells.

Methods

The cells were seeded in 96-well plates at 1×105 cells/well and treated with 10-150 μm of the 2, 4-diamino-1, 3-diazaspiro [5.5]-9-tert-butyl-2, 4-diene- 5- carbonitril (9-tbap). this compound was found to be highly active cell growth inhibitor with ic50 of 30±3.5 μm as determined by mtt assay. evaluation of survivin expression in nb4 cells treated with 9-tbap was performed by real time pcr.

Results

9-tbap decreases cell proliferation of the nb4 cells in a dose- and time-dependent manner. the ic50 value following 72 h exposure was found to be 30 μm for the cells. furthermore, real time pcr analysis revealed that the treatment with the compound down-regulated the expression of survivin in a time dependent manner.

Conclusion

: these data further suggest that 9-tbap may provide a novel therapeutic approach for the treatment of leukemia.
