Introduction: Leukemia is a clonal malignancy of the myeloid or lymphoid progenitors in the bone marrow. Acute myeloid leukemia (AML) progressed from somatic mutations in myeloid lineage precursors coupled with transcriptomic dysregulation of bone-marrow infiltration that lead to the production of immature myeloid cells (blasts) and disruption in normal haematopoiesis(1). In this study, we predict the miRNAs and genes in bone marrow mesenchymal stromal cells using bioinformatics software and GEO dataset.
Methods: Using the GEO database (GSE122917) and analyzing the expression profiles of genes of AML patients (bone marrow mesenchymal stromal cells) genes were examined and finally KLK8 gene selected. After getting the KLK8 protein chain from the NCBI database, the miRNAs targeting the gene were predicted using TargetScan, miRDB, and miRWalk databases using different algorithms.
Results: Based on the scoring system of the bioinformatics programs and considering the best targeting scores, hsa-miR-6820-5p, hsa-miR-6080, hsa-miR-6882-3p, hsa-miR-1225-3p were identified as miRNAs targeting KLK8 gene . They were suggested for the practical studies in future.
Conclusion: Results from bioinformatics studies revealed that of the four miRNAs identified and are probably the targeting miRNAs specific for the KLK8 gene , respectively . therefore it seems that these miRNAs can be a suitable candidate for in vitro studies in AML patients
